If the combo is NAIL-SPINE, I think that the clue would be pointing us to the spine in your body, and LOCK would involve metal. Being that it's the opening clue and it's a 2, I think that it's more likely to be the first-instinct, obvious combo. On a 3 or 4, I agree those would qualify.

Nail was my first thought. Card/Spine/Lock all fit the clue as a second. I'm just trying to think where OH is trying to lead us. I don't like Spine or otherwise the clue should be different (directing to anatomy), same with Lock + Nail you surely wouldn't clue with File. So Card is probably the strongest.
