Huh. I've never been able to get the Earthmother to work right. And, yes, if you do it properly you can get it at level 2, which would be absurd. For the Transmuter, who doesn't care about the Earthmother's penalty for having Negative Piety, there's no reason not to go all-out and blast every wall you can once you've got Stoneform.

I never thought of that method (go level up in the middle of a boss fight for the refills), but saw it on the DD wiki. It seems a really exploitative loophole of the game mechanics, but it sure is effective. And now I wonder if you could pull off that XP catapult for other purposes - like use the 3/4 level up to kill a level 7 or 8 monster.
Overall I mostly like the balance between the fireball and physical attacking. It's rarely either-or; usually the best answer is a combination of them. I think the missing part is that the wizardly classes should be able to do more damage with a fireball, not just cast more fireballs. It seems silly that even a berserker gets good mileage out of the spell.

The biggest problem with wizardly classes is not finding the glyphs you need. I recently played a game where I explored 70% of the dungeon before I found a single glyph -- and that was the wall destroying one.  Did not find the fireball until the dungeon was 90% explored.  Lots of mana, never used. At least I could take on mana-burning enemies with no worries.
Turned that expedition into a "oh well at least I gathered some gold for my next real game" trip.

The real king of the fireballs, though, is the vampire class, since he can use his HP to cast spells.

Ha, I somehow didn't realize that the advanced glyphs get unlocked for normal play. I thought they were exclusive to their classes (healing for Paladin, etc) and was only playing the challenge dungeons. So the game just got rather easier.  The poison glyph in particular is rather overpowered; with it, anything you can survive a hit from you can eventually kill. A Monk with poison is particularly diesel, with his phys resistance and double regen.
An Assassin in the Crypt is hard? Try a GORGON in the crypt, with the same poison reliance oh and -50% melee damage thrown in. I actually pulled that off, though only by resorting to the polymorph god. (which itself took several goes to get right - needed to get the altar literally in the starting field of view.)

I still haven't made Mystera work at all consistently. I guess you really need to totally dedicatedly spam a cheap spell beforehand, and the only two that are suitable are Bysseps and Halpmeh. (or Endiswal with the transmuter.) Problem is that Mystera really needs both the +15 max mana and +25% fireball damage before you get anywhere useful, but that 70 piety total takes forever to rack up. Not to mention the opportunity cost of waiting for Mystera if one of the more straightforward deities (Pactmaker, Jehora Jeheyu, Tikki Tooki) is also present.

Mystera can be decent with BLUDTOPOWAH, it seems tailored for her. Without that, I don't see any way for her to be better than 4th or 5th pick. If you use the Stoning bug you can get to max piety in no time, though.
